| IL-23

Interleukin-23 is a heterodimeric cytokine composed of the IL-12B (IL-12p40) subunit (shared with IL-12) and the IL-23A (IL-23p19) subunit. The functional receptor for IL-23 (IL-23R) has been identified, consisting of IL-12R β1 and IL-23R. Adnectin-2 binds to IL-23 and competes with IL-23/IL-23R. The IL-23R exhibits some single nucleotide polymorphisms in its IL-23-binding domain, potentially leading to variations in Th17 activation. IL-23 has been demonstrated to be a key cytokine for Th17 maintenance and expansion. IL-6 and TGF-β activate the Th17 transcription factor RORγt, inducing Th17 polarization. IL-23 stabilizes RORγt, enabling Th17 cells to function properly and release their effector cytokines—such as IL-17, IL-21, IL-22, and GM-CSF—which mediate protection against extracellular pathogens (fungi and bacteria) and participate in barrier immunity.
